HOW DO POLYNUCLEOTIDES WORK?
Polynucleotide molecules are injected into the skin where they interact directly with fibroblast cells — the cells responsible for producing collagen, elastin, and hyaluronic acid. The DNA fragments activate specific fibroblast genes that control collagen synthesis, elastin production, and tissue repair. This is regeneration at a genetic level.
At the same time, polynucleotides stimulate angiogenesis — the formation of new blood vessels — improving oxygen and nutrient supply to the skin. They also inhibit melanogenesis (reducing pigmentation), provide potent anti-inflammatory and antioxidant protection, and induce accelerated cell turnover.
The process happens in stages:
Stage 1
Immediate (days 1–7)
The treatment area may feel slightly firmer and look mildly improved due to the initial inflammatory response triggering early fibroblast activity. Mild redness and small raised blebs are normal and resolve within hours to a couple of days.
Stage 2
Collagen Stimulation (weeks 2–6)

WHAT DOES A POLYNUCLEOTIDE TREATMENT INVOLVE AT SEBASTIAN ROSE?

Your Treatment Plan
Most patients require a course of 3 sessions spaced an average of 3 weeks apart. Patients combining polynucleotides with skin boosters follow a 4-session protocol — 2 PN and 2 skin booster sessions, all 14 days apart. The right protocol for you is determined at consultation.
The Treatment
Sessions take 15–30 minutes depending on the areas treated. A topical numbing cream is applied for comfort. The polynucleotide solution is injected using a fine needle with a micro-papule or linear threading technique depending on the area and formulation. The treatment is well-tolerated and discomfort is minimal.
After Your Treatment
You may experience mild redness, small raised blebs, and a sense of warmth in the treated area immediately after treatment. These resolve within hours to a couple of days. Avoid touching the treated area, wearing makeup for at least one hour (ideally six hours), and avoid saunas, steam rooms, and intense exercise for 48 hours. Apply SPF 50 daily.
Follow-Up
We ask all polynucleotide patients to return approximately 20 days after each session so we can assess skin progress and ensure the protocol is on track.

PDRN (polydeoxyribonucleotide) is the specific DNA fraction used in polynucleotide treatments. The terms are often used interchangeably. All formulations we use are highly purified PDRN-based injectables. We offer both injectable polynucleotides. and in conjunction with microneedling, we prefer to use a PDRN formulation.
Can polynucleotides treat hair loss?
Yes. Polynucleotides are an effective treatment for androgenetic alopecia and diffuse hair thinning when injected into the scalp. They stimulate follicle activity and improve scalp tissue health. A course of 3-4 sessions is recommended.
